to rent a car at the San Jose Del Cabo airport in Mexico, About how long would it take to drive to La Paz.

It's 137 miles on crappy two-lane road and you'll have plenty of company. I did the same but to Todos Los Santos - worth the trip but it's not like a grocery-store run in the States. So, I'd give yourself 3 hours.

I would suggest keeping some cash on you in case you have to pay a traffic "fine" if you get pulled over.
Be sure to get the insurance. We did the Todos los Santos to La Paz loop a few years ago, parked on a desolate (or so we thought) Pacific side beach near Todos Santos, took a long walk on the beach, and came back to a busted out rear passenger window. Nothing taken (anything good was in the trunk). Fortunately I had a roll of duct tape along to seal up the fish cooler for the trip home, so I constructed a duct tape window, and we drove around for a few more days in the La Paz area with glass all over the back seat. When we arrived back at the airport in San Jose del Cabo the rental check-in crew was very excited, then very disappointed when I told them "no problem, full coverage". Other than that, it was a great trip.

Cows are a bigger threat than crooked cops or bandidos
So true, the cows are ridiculous. I sat in a 10 car lineup on the road back from Todos Santos trailing a cow on the road. Probably why there are little white flower adorned shrines on the side of the road every 50 yards.
